Description

KPV (Lys-Pro-Val) is a synthetic tripeptide corresponding to the C-terminal sequence of alpha-melanocyte-stimulating hormone (α-MSH) [5]. As a member of the melanocortin-derived peptide family, it retains structural features associated with the parent hormone while lacking the pigment-inducing activity characteristic of full-length α-MSH [3]. Its compact three-residue architecture — lysine, proline, and valine — has made it a widely employed tool in peptide biochemistry and receptor-interaction studies.

A key structural attribute of KPV is its recognition by PepT1, the intestinal di/tripeptide transporter that is normally expressed in the small intestine and upregulated in colonic epithelial tissue during inflammatory conditions [2]. Researchers exploit this transporter relationship to investigate how small peptides are taken up into epithelial and immune cells, and to probe downstream signaling events within those cellular compartments.

Laboratory investigations have used KPV as a model compound to examine NF-κB and MAP kinase signaling in the context of inflammatory cytokine regulation [2]. Its structural relationship to α-MSH also positions it as a reference scaffold for researchers studying melanocortin receptor biology, mucosal epithelial responses, and dermal cell behavior in vitro and in preclinical models [3]. Additionally, KPV-containing sequences have been incorporated into larger peptide frameworks to explore structure-activity relationships in antimicrobial and antifungal contexts [1].
